
This is a biotherapeutic project focusing on the production of inactivated vaccines. It covers the entire production process from pathogen culture, inactivation, purification to formulation, filling, and packaging. The project is equipped with a GMP-compliant production system that meets the strict guidelines of the FDA and EMA, integrating state-of-the-art professional cleanrooms, specialized biosafety protection equipment and high-efficiency bioprocess equipment to ensure product quality, biosafety, and regulatory compliance.
Key Challenges: During the project implementation, the main challenges we encountered were focused on the plant design and production operation for large-scale inactivated vaccine production, specifically in the following three aspects:
1. Biosafety: Inactivated vaccine production involves live pathogens in the early culture stage. Even after inactivation treatment, strict biosafety control is still required to prevent the leakage of live pathogens or inactivated antigenic substances, which poses high requirements for the plant’s sealing performance, air circulation control, and waste treatment, bringing great challenges to biosafety management.
2. Personnel protection: The production process involves live pathogens and potentially harmful substances (such as inactivation reagents). Operators are at risk of exposure to pathogens or toxic reagents during operation, requiring comprehensive and strict personnel protection measures, including the configuration of professional protective equipment, the setting of scientific decontamination procedures, and the division of safe operation zones, which increases the difficulty of personnel management and operation standardization.
3. Prevention of product contamination: Inactivated vaccines have extremely high requirements for product purity and sterility. The production process is prone to contamination by microorganisms, foreign impurities, or cross-contamination between different vaccine batches, requiring strict control over raw materials, production equipment, clean environments, and operation processes to ensure the safety and effectiveness of the final product.
Targeted Solutions: To effectively address the above challenges, we customized targeted and feasible solutions to ensure the project’s smooth progress, stable quality, and full compliance with regulatory requirements:
1. Strengthened biosafety control measures: We adopted a hierarchical biosafety design for the plant, strictly divided biosafety zones (such as BSL-2 or BSL-3 zones according to pathogen risk level), configured high-tightness isolation equipment and negative pressure ventilation systems to prevent pathogen leakage; established a full-process biosafety monitoring system, set up multiple monitoring points in key production links, and formulated strict disinfection and decontamination procedures for the plant, equipment and environment to ensure comprehensive biosafety control.
2. Comprehensive personnel protection solutions: We formulated a customized personnel protection plan, divided the plant into different levels of operation zones, configured professional protective equipment (PPE) suitable for different operation links (such as full-body protective suits, gas masks), set up dedicated decontamination channels and buffer zones for personnel entry and exit, and carried out regular professional training for operators on protective operation specifications and emergency response, ensuring the safety and health of personnel.
3. Multi-level product contamination prevention measures: We established a full-process contamination control system, strictly inspected and qualified raw materials before use; selected sterile production equipment and closed transfer systems to avoid contact between products and external pollutants; optimized the cleanroom design, maintained a high-level sterile environment, formulated strict operation specifications, and implemented batch separation management to prevent cross-contamination between batches, ensuring the sterility and purity of the final vaccine product.
